Assay principles:
- Biotinylated recombinant thermostable luciferase can via streptavidin be linked to any biotinylated primary or secondary antibody. After washing and addition of luciferin and ATP, luminescence output can be immediately recorded in e.g. a microplate luminometer.
- Extremely high sensitivity is obtained by using biotinylated acetate kinase. ADP + acetyl phosphate --------> acetate + ATP Bioluminescent detection of the ATP by the firefly luciferase enzyme can be used to detect the marker enzyme down to 10-21 moles/assay.
