Basic research of immunological mechanisms for chronic graft dysfunction

As one landmark in modern medicine, organ transplantation has brought new lives to a large number of end-stage patients. However, the development of organ transplantation is challenged by two main obstacles: the severe shortage of donor organ and Chronic Graft Dysfunction (CGD), the chronic irreversible function decrease in the allograft. Studies in China and western countries on CGD have lasted for more than 30 years, but the exact mechanism remains unknown, and the intervention to CGD is still poorly developed. This project focuses on the following research topics: 1. The mechanism and intervention of hepatitis B relapse after liver transplantation for HBV-associated diseases; 2. The immuno-regulative mechanism of small molecules on CGD; 3.Evaluation and application of new methods for selective negative- regulation in the prevention of allotransplantation rejection; 4. The immunological mechanism of CGD; 5. Identification and application of early diagnostic parameters for CGD; 6. The relationship of between microecosystem imbalance and CGD after organ transplantation; 7. The molecular mechanism and intervention of tumor recurrence after organ transplantation; 8. Design of novel immunosuppressor for CGD and establish high-throughput screening system; 9. Establish human leucocyte antigen (HLA) three-dimensional model and its evaluation system in liver transplantation. Successful studies will clarify the molecular mechanisms of CGD, hepatitis B relapse as well as tumor recurrence after organ transplantation, which will greatly benefit the early diagnosis of CGD and development of effective intervention approaches. And it will also broaden our knowledge about the regulation of immunoreactions in autoimmunity, infection immunity and tumor immunity.
