Differentiation of human embryonic germ cells and the plasticityof adult stem cells

Stem Cell is a primitive cell with self-renewing ability and multi-differentiation potential. Under specific condition, Stem Cell could be coaxed into different cells, constructed different tissues and organs. At present, different kinds of cytosine are used to learn the mechanism of stem cell differentiation, but the method could not lead to the systematic information of differentiation mechanism. In our project, we used the IVF technique to get human embryonic stem cell, comparing several feeder layers, and constructing the first embryonic stem cell line of China. On the basis of stem cell line, we will construct cell models about the embryonic stem cell differentiating into neuron, and the stromal stem cell and neural stem cell differentiating into other function cells, then compare systematically the protein expressed between embryonic stem cell and neural stem cell, neural stem cell and other adult stem cell, find and clone membrane proteins and secret proteins which could be the key proteins in regulating the stem cell differentiation, and make the mono-clone antibodies of those proteins. Furthermore, we make research on the function and the mechanism beneath those proteins in regulating stem cell differentiation, and the relationship between different signal pathways. This project would provide a theoretic basis for curing diseases with stem cell.
