
Science:小鼠试验提出一个生产可移植细胞的方法
生物谷报道:最近,在一项用小鼠做的试验中,研究人员分离出通过一个叫“孤雌生殖”的过程生产的胚胎干细胞。这一研究结果发表在最新一期的《science》杂志上。
孤雌生殖指从未受精卵开始发育的胚胎,由于这个过程发育的胚胎有母系基因组的两个拷贝,而不是一个父系和一个母系基因组拷贝,它们应该与卵子的供体有更具体的匹配。尤其能使细胞表面名为MHC的抗原紧密匹配,这些蛋白在每个个体几乎都是唯一的。当一个外源物质进入身体时,MHC蛋白成为免疫系统T细胞的靶标,所以MHC的不匹配是移植的器官和组织被排斥的一个主要原因。
Kitai Kim和同事用来自有丝分裂两个不同阶段的小鼠卵细胞诱导了孤雌生殖发育,并且从由此得到的胚胎中分离出干细胞,这些干细胞带有卵子供体全部的MHC蛋白。这些干细胞能产生许多、但不是全部的组织,这也许与缺少父系印记的基因组有关。这个方法需要大量的卵细胞,是否能用于人类还有待研究。
