生物谷报道:美国Roswell Park研究中心的科研人员在肿瘤综述杂志上提出新观点,维生素D信号通路是潜在的抗肿瘤治疗途径之一。
维生素D代谢通路:25(OH)D3被运至肾皮质部,被肾脏的25-羟基维生素D3 1α-羟化酶和24-羟化酶分别羟化为1,25(OH)2D3 和24,25(OH)2D3 。1,25(OH)2D3是生物活性最强的代谢物。
流行病学调查发现,人类多种肿瘤的病因与维生素D缺乏有关。临床前期研究也发现,活性维生素D(α25(OH)(2)D(3),钙三醇)及维生素D类似物有抗肿瘤作用,抑制肿瘤细胞增殖,促进其调亡,及抑制血管生成。此外,钙三醇抗肿瘤效应还影响多种细胞毒素和抗肿瘤药物。
由此,研究者总结认为,从流行病学、试验及临床研究,都支持维生素D及维生素D类似物具有防治肿瘤的作用。
