及表达强度的变化,初步阐明两基因对MSCs分化为心肌细胞的影响。以MSCs植入后的Wistar大鼠为研究对象,在移植后不同的时间点取心肌标本,采用逆转录聚合酶链反应技术,提取心肌组织的总RNA ,逆转录,加入特异性引物扩增目的基因,观察基因表达的时序性,并应用凝胶图像分析系统检测这两个基因表达强度的动态变化。显示在MSCs移植后1d两个基因即出现弱的表达,1d~1周表达较低,2周~3周表达强度最高,达到高峰期,以后表达强度则逐渐降低。NKx2.5、GATA 4在MSCs转化为心肌细胞的早期发挥重要的作用
To investigate expression and quantity of Nkx2.5 and GATA-4 during MSCs differentiation into cardiomyocytes and to illustrate its effect on differentiation process. Experiments were performed in 18 Wistar rats after MSCs transplantation. At intervals myocardial specimens were obtained after MSCs transplantation, total RNA was acquired, reverse-transcripted and added into special primers and objective gene were expanded by PT-PCR. We observed the timepoint of gene express initially, the change of gene quantity was detected by erodes-photo analysis system. Nkx2.5 and GATA-4 expressed low one day after MSCs transplantation, its expression were up to peak at 2 week~3 week and from then on diseased. Conclusion: NKx2.5、GATA-4 played an important role on early-phase of MSCs differentiated into cardiomyocytes in vivo.
